Hello, I am using the DB Pro plugin for a while now and it’s working great. However, we are noticing a lot of spam comments, so I decided to add the user login/registration to require users who want to comment to register/login before doing so. I have read the documentation several times and am having no luck getting this setup correctly.
This is a private site, so I’ll need to provide you with a separate login not posted on this forum. Can you please assist?

If it's a private site, I don't understand how you can be getting spam comments - unless it's possible for users to access the site?
To ensure that only registered users can post or comment, ensure that you have selected permitted poster roles in Discussion Board > Settings > User tab.
